Gareth Bale is set to start for the Spaniards after returning in Saturday's win at Sevilla following a calf injury.
Juve pair Andrea Pirlo and Giorgio Chiellini are expected to play.
The veteran Italy internationals were both dropped to the bench as the Bianconeri won 1-0 at Sampdoria to clinch their fourth successive Serie A title.
Pirlo, 35, is set to renew acquaintances with Real Madrid boss Carlo Ancelotti, who managed the midfielder during a successful eight-year spell at AC Milan.
The pair helped the Rossoneri win a Serie A title, two Champions Leagues and the Club World Cup between 2001 and 2009, before Ancelotti left to take over at Chelsea.
"He's like a father to me," said Pirlo. "We worked together for many years, he gave me my chance in top-level football and changed my position.
"We won a lot together and I experienced unforgettable moments with him."

Ahead of the game, Pirlo described Real as the "most important club side in the world".No team has made as many European Cup (now the Champions League) final appearances than Real's 13 and no other team has won as many titles as the Spanish giants, who claimed the trophy for a 10th time last season.
Pirlo, however, says the Italians will not be overawed.
"The first leg is crucial. We'll need to play with intelligence," he said. "We'll be mindful but also show some swagger, because we have a dream to fulfil and nothing to lose."
